      • Duplessis wurde in eine Familie mit künstlerischen Neigungen geboren. Sein Vater, ein Chirurg namens Joseph-Guillaume Duplessis, gab seinen Beruf auf, um sich der Malerei zu widmen und ihm anfängliche Schulungen zu geben.
      • Er studierte unter Joseph Gabriel Imbert (1666–1749), einem Schüler von Charles Le Brun, und erwarb grundlegende Fähigkeiten in der Maltechnik.

      • Er trat der Académie de Saint-Luc bei und begann, Porträts auszustellen, obwohl die anfängliche Anerkennung begrenzt war.
      • Ein bedeutender Durchbruch gelang ihm beim Pariser Salon von 1769, wo seine Ausstellung von zehn Gemälden großes Lob erntete, insbesondere von Denis Diderot.
